   <subfield code="a">The distribution of actin microfilaments in two cell lines, ras/mycSFME and r/mHM-SFME-1, was studied under colony-forming conditions in soft agar gel. Both cell lines were transformed by human c-Ha-ras and mouse c-myc genes. The r/mHM-SFME-1 cell line, which is derived from ras/mycSFME, shows highly metastatic potential in host animals, whereas ras/mycSFME shows low potential. Colonies of the r/mHM-SFME-1 cell consisted of loosely packed cells with dispersed satellite cells around the colony, whereas the ras/mycSFME cells produced fairly compacted colonies. The distribution of actin microfilaments in the submembranous region, especially at the cell-cell adhesion site, was more frequent in the ras/mycSFME cells than that of the highly metastatic r/mHM-SFME-1 cells. The cell-cell adherens junction developed well in the ras/mycSFME cells as compared to that of the r/mHM-SFME-1 cells. When these cell lines were cultured on fibronectin-coated dishes, however, both expressed a fibroblastic appearance and spread well over the dishes. Therefore, the phenotypic differences in the metastatic potentials of the two cell lines were demonstrated by the three-dimensional cultures in soft agar gel.</subfield>
