   <subfield code="a">Summary: Recent studies have demonstrated significant skin and RAST reactivity to spore extracts of multiple basidiomycete (BS) species in some atopic subjects. The association of RAST reactivity to different basidiomycete species suggested the presence of shared allergenic determinants. This was confirmed by RAST inhibition studies with spore extracts from different basidiomycete species. Preliminary investigations suggest, however, that basidiospore allergens do not share extensive cross-reactivity with members of the Fungi Imperfecti. Immunoprint analysis of proteins fromCalvatia cyathiformis, separated by isoelectrofocusing, demonstrated a number of protein bands that bound IgE from RAST positive sera. IEF/immunoprinting revealed three allergen(s)/groups or bands (Bd) with respective pI of 3.6-4.6, 6.6 and 9.3. Only Bds 3.6-4.6 were stable at 37°C or at acid pH. Bd 9.3, the most labile, reacted with 63% of the patients' sera tested. Immunoprint inhibition studies of 6 BS species suggest that Bd 9.3 fromCalvatia cyathiformis is a common allergen in other basidiomycetes. Purified fractions ofCal c Bd 9.3 were obtained through preparative isoelectrofocusing, gel filtration, and hydrophobic interaction chromatography. These studies demonstrate that basidiospores are important fungal aeroallergens that share allergenic determinants. Further efforts are directed at isolation and characterization of major shared basidiospore allergens.</subfield>
